Tissue organ bath system – 750TOBS

Organ baths tend to occupy a lot of bench space, experience temperature delays and gradients with circulating water in the double-jacketed components, are labour-intensive to clean, are difficult to automate, and for the most part, are non GLP compliant. To remedy these limitations, DMT has redesigned the organ bath system for the new millennium using the latest developments in material and computer and tissue engineering:

  • took away the circulating water-heating and replaced it with rapid air-heating for more efficient and consistent temperature control
  • took away the double-jacketed glass chambers and replaced them with single-wall glass chambers to make cleaning easier
  • added computer-automated filling and emptying control to ease workload
  • provided digital force output for higher quality data acquisition
  • managed to put all of this in a box that takes up a quarter of the space of a traditional system.

The modular design allows up to four individually mounted tissues to be studied simultaneously and independently per system. Moreover, multiple systems can be linked together (via USB connection) for high throughput screening which can be easily operated by a single person. The convenient, self-contained, compact bench-top design of DMT’s 750TOBS can be placed in any existing laboratory situation.

The 750TOBS has been used extensively to investigate the physiology and pharmacology of smooth muscle and other muscular tissue preparations. These studies offer the advantage that preparations are subjected to pharmacological agents without the influence of systemic processes present in intact animals. The data of these organ bath systems is generally more consistent and reproducible and serves as a tool for tissue screening and for pharmacology studies (i.e. concentration response curves).

The most common application of DMT tissue organ bath:

  • Cardiovascular research, using isolated aortic rings, heart tissue (papillary muscle, left ventricle) or other muscle strips.
  • For gastro-intestinal research preparations of ileum and colon are often used, as well as gastric antral muscle and sphincter.
  • Respiratory physiology and pharmacology can be studied in isolated phrenic diaphragm preparations, pulmonary arterial smooth muscle and even lung parenchyma.
  • Other smooth muscle preparations that are used in organ bath research include urinary bladder and penile muscle strips.
